Characteristics of the patient specimen from which SUM-102 was derived

The SUM-102 cell line was derived from the mastectomy specimen of a 57 year old patient who had been diagnosed with locally advanced breast cancer and had been previously treated with chemotherapy.  Histopathological analysis of the mastectomy specimen revealed an unusual presentation, considering that the patient was thought to have had locally advanced breast cancer.  The pathology report indicated a minimally invasive apocrine type adenocarcinoma arising in a papillary lesion, possibly a remote intraductal papilloma with abundant unusual intraductal proliferation, possibly intraductal apocrine carcinoma.  14 lymph nodes examined were negative for cancer, and the cells were negative for estrogen and progesterone receptor.

After seeding cells from this specimen into culture, we found that the medium that best supported their proliferation was Ham’s F12 medium supplemented with 5% FBS, insulin, hydrocortisone and epidermal growth factor (EGF).  Indeed, these cells were/are dependent on exogenous EGF for growth in culture.
