The HCC70 Breast Cancer Cell Line

The HCC70 human breast cancer cell line was derived from the primary tumor of a 49 year old African American patient diagnosed with breast ductal carcinoma.  The cell line is ER and HER2 negative and has been classified as a Basal A type breast cancer.  The STR profile along complete genomic information for this cell line can be found at Cellosaurus, at COSMIC, or at DepMap Portal.

Oncogene Signature:

The Oncogene Signature of this cell line is fairly typical for a Basal A type breast cancer cell line.  It should be noted here that, at this time, there is some discrepancy between the oncogene signature that we derived from our database and that found in the Expasy data base.  As shown below, these cells have a PTEN mutation and RB1 mutation.  In addition, the Expasy database indicates that these cells also have a TP53 p.Arg248Gln point mutation, which again, would be typical for a Basal A breast cancer cell line.  The cells have an amplification  and high level overexpression of the CDKN2B gene, but this gene was not a screen hit.  Given that PTEN is mutated in this line, it is also of interest that RICTOR is amplified and overexpressed, though not a screen hit.

HCC70 Drug sensitivities.  Despite the fact that this is a Basal A cell line which rarely have functional-druggable oncogenes, the cell line does present with a number of interesting drug sensitivities many of which seem to be related to the PTEN status of this cell line and the resulting importance of PI3’K-AKT signaling in this cell line.  PIK3CB was a screen hit and the cells are sensitive to a cognate targeted drug.  In Tier 3, there are several other drugs to which the cells are highly sensitive that target one or more AKT family members.  The cells are also quite sensitive to several pan-specific drugs that target cell survival regulators, WNT signaling regulators and others.  Thus, as a Basal type breast cancer cell lines, there are several ways to attack these cells using low doses of several targeted drugs, the most important of which are connected to the PTEN status as shown in the Oncogene Signature.
