Derivation of the SUM-185PE cell line

This cell line was derived from a 700 ml pleural effusion specimen that was procured in the lab on November 29th, 1995.  Cytology analysis of this specimen was positive adenocarcinoma cells consistent with a breast primary.  By this point in the development of the SUM lines, we knew that the Ham’s F12 medium supplemented with 5% FBS, insulin and hydrocortisone was our best medium for initiating these cell cultures.  The SUM-185PE cells did well in this medium from the very beginning.  The cells were maintained in primary culture for approximately six weeks, which was not at all unusual, and the first passage was accomplished by mechanical agitation of the culture dishes to dislodge cells piling up in colonies.  The first use of trypsin:EDTA came in the second passage, which occurred in late February of 1996.  Thereafter, cells were split with a 1:3 ratio approximately every two to three weeks.
