This tool allows users to determine, in rank order, the most essential pathway in each breast cancer cell line, or alternatively, to choose any KEGG pathway and see the rank order of cell lines that rely on that pathway. The “essentialness” for each cell line and pathway is determined using an algorithm that considers the fractional enrichment of a pathway with genes that were hits in the shRNA screen, and the hit genes are weighted if they are directly connected in the pathway with other hit genes, and weighted further if they are genomically altered. By using this tool, users can toggle between the KEGG Pathway Analysis tool and the KEGG pathway Engine to see how the essentialness scores translate into screen hits in the pathway, and the druggability of the pathway.
